The makers of the film have researched what foods humans have thrived and developed on throughout the ages. They certainly make a strong case for the notion that we are not best suited for a vegan or vegetarian diet, as the eras when our brains developed were meat eating times. I am not sure that they make a great case for eliminating grain, however. Just because humans did not eat them does not necessarily mean they are bad for you. They do show some evidence that eating grains can lead to weight problems, or more aptly, that eliminating them can lead to fat loss.
